p97 ATPaseを利用したユビキチン非依存的なタンパク質分解システムの開発
Abstract
本研究では、p97 ATPaseを利用した新規のプロテアソーム分解システム(p97 PROTAC)を開発した。p97 PROTACは、p97アダプター蛋白質のUBXドメインとナノボディを融合させた人工的なアダプターで、ターゲットタンパク質を p97に結合させ、プロテアソームによる分解を誘導する。
具体的には以下の知見が得られた:
p97 PROTACは、細胞内の様々な局在を持つタンパク質(核内、核膜、転写因子など)を効率的に分解できる。
p97 PROTACは、53BP1などのリキッド-リキッド相分離構造に局在するタンパク質の分解も可能である。
p97は神経変性疾患の病態に関与する凝集体の分解にも利用できる可能性がある。p97は脳内の関心領域(大脳皮質、海馬、黒質)に広く発現している。
p97 PROTACによる分解は、ユビキチン化非依存的に進行する。p97の ATPase活性は必須ではない。
以上より、p97 PROTACは従来のユビキチンリガーゼ依存的なPROTACとは異なる新しい分解システムであり、大型複合体や膜タンパク質、凝集体の分解に有用な可能性がある。

p97 PROTAC (UBX-Nb(GFP))は、GFP融合タンパク質(GFP-Coilin、GFP-Emerin、GFP-ETV1)の発現を有意に減少させた。
p97 PROTAC (UBX-Nb(GFP))は、53BP1-YFPノックインU2OS細胞において、53BP1の発現を有意に減少させた。
p97は、マウス、ラット、サルの脳の大脳皮質、海馬、黒質線条体に広く発現している。
